Abstract Background The molecular characterization information of T-DNA integration is not only required by public risk assessors and regulators, but also closely related to the expression exogenous endogenous genes. At present, with development sequencing technology, whole-genome resequencing has become an attractive approach identify unknown genetically modified events characterise events. Results In this study, we performed genome Pb29, a transgenic high-resistance poplar 741 line that been commercialized, using next-generation Nanopore sequencing. results revealed there are two insertion sites, located at 9,283,905–9,283,937 bp on chromosome 3 (Chr03) 10,868,777–10,868,803 Chr10. accuracy locations directions was verified polymerase chain reaction amplification. Through sequence alignment, different degrees base deletions were detected left right border sequences, in flanking sequences sites. An fragment inserted between Chr03 site sequence, Pb29 did undergo chromosomal rearrangement. It worth noting detect API gene genome, indicating containing BtCry1AC gene.
